AOD-9604 is a synthetic peptide fragment derived from the C-terminal region of human growth hormone, corresponding to amino acids 176 through 191 of the hGH sequence. The peptide consists of 16 amino acids. It has a molecular formula of C78H123N23O23S2 and a molecular weight of 1815.1 g/mol. Structurally, AOD-9604 contains an additional tyrosine residue at the N-terminal end to stabilize the peptide and is cyclized through disulfide bonding between two cysteine residues.
Researchers at Monash University in Melbourne, Australia developed the peptide during the 1990s. The designation “AOD” represents “anti-obesity drug” and reflects its original development intent as an obesity treatment. Researchers at Monash University, including Frank M. Ng and colleagues, broke down whether the lipolytic activity of full-length human growth hormone could be isolated from its insulin-antagonizing effects. The rationale to use a C-terminal fragment of hGH lies in its physiological function, which includes stimulation of lipolysis in adipocytes.
